You can unlock new Heroes by collecting their Hero Fragments. You can view the list of unlockable heroes by selecting the "Heroes" tab on the bottom right of your screen. There, you can see this list of available heroes and how many Hero Fragments you need to unlock them. 

You can also instantly unlock Heroes by purchasing Hero Crates.

Additionally, there are some Heroes that are unlocked as you progress through the different districts.
